Microscopic identification

Root cross-section: The cork layer consists of a dozen layers of yellow-brown cork cells; the cortex is thin, consisting of parenchyma cells; phloem ray cells are in 2-3 rows; most of the fiber bundles are in the cortex and phloem; The tangential extension is about 5-6 layers; the xylem is broad and the ducts are arranged radially. The parenchyma in the cortex and phloem contained many starch grains.

Powder characteristics

Yellow white. 1 Cork cells yellow-brown. 2 The phloem parenchyma cells are round or irregular and have cell gaps. 3 mesh catheter, occasionally bordered pit catheter, diameter 30-50μm. 4 fibers are colorless and 7-15μm wide. 5 starch grains are mostly units, round, helmet-shaped hat, striae are not obvious, umbilical point-like or crack-like, diameter 3-15μm.

Physical and chemical identification

Take this product coarse powder 5g, add ethanol 20ml, set on water bath reflux 1h. After filtration, the filtrate was concentrated to 5 ml for the following experiment:

(1) Take a test solution 1ml, add a little magnesium powder, a few drops of hydrochloric acid, set in a water bath and heat for a few minutes, place a magenta color.

(2) Take 1ml of the test solution, evaporate the evaporating dish and dry it, add 1ml of boric acid saturated acetone solution and 10% citric acid acetone test solution, continue to evaporate and observe under the ultraviolet light, and show yellow fluorescence.

(3) Take 1 drop of test solution on the filter paper, spray 1% aluminum trichloride ethanol solution to dry it, observe under the ultraviolet light, and show yellow fluorescence.
